Kwame Nana's late winner takes the hosts to the 10th position with 27 points from 20 games while the visitors are still rooted to the bottom of the league table.
Nsoatreman FC took the lead through Samuel Ofori, who converted a penalty in the 32nd minute to give the home side a 1-0 lead.
Kotoku Royals struck back nine minutes later, with Collins Kudjoe the scoring to level the game up for the visitors.
The first half ended in 1-1 as no more goals were scored in the remaining four minutes.
In stoppage time, Kwame Nana stole all three points for the home side at the expense of a vulnerable Kotoku Royals.
Nsoatreman FC will travel to Ainyinase to play as guests to Karela United in week 21 of the 2022/23 Ghana Premier League on Sunday, March 12, 2023.
For the visitors, they will host Bechem United the same day.
